The oligonucleotide must be split from the solid support and the protective groups must be removed before it can be employed in downstream applications.

OLIGOS PRODUCTION PROCESS

The demand for oligonucleotide manufacturing has increased dramatically in recent years. The oligos are

generated using a solid-phase synthesis, which results in a higher yield of purified product, which is subsequently used in polymerase chain reaction (PCR), DNA sequencing, library construction, and artificial gene synthesis. The oligonucleotide must be split from the solid support and the protective groups must be removed before it can be employed in downstream applications. The synthesis procedure is now automated and computer-controlled, but the post-processing, cleavage, and deprotection steps are still done by hand. This method causes a bottleneck in the production workflow, reducing productivity and efficiency. To address this challenge, Milestone created wavePREP: an efficient, quick, and safe solution for the oligos post-processing stage. Its approach combines cleavage and deprotection into a single step, ensuring a superior workflow as well as higher product yields by eliminating the evaporation stage.

CLEAVAGE AND DEPROTECTION STEPS

Following the completion of the synthesis procedure, the oligos must be taken from the solid support and deprotected. Because these manual stages take several hours to complete, they have a direct impact on the ultimate yield, as well as on productivity and the lab’s workflow. These procedures are typically carried out by immersing the plates in ammonia solutions or other chemicals, necessitating an evaporation phase and so creating unpleasant lab conditions. WavePREP takes a novel approach to post-synthesis by combining oligo cleavage and deprotection in a single step and eliminating evaporation.

In wavePREP, liquid reagents convert to gas while simultaneously cleaving and deprotecting oligos. To achieve perfect process control and maximise yield, reaction conditions are precisely controlled. Following completion of the operation, oligonucleotides can be directly desalted, for example, using acetonitrile, and eluted from the solid support with the appropriate buffer solution.
